Determinación del potencial celulolítico de bacterias asociadas a compost de estiércol bovino y pasto

dc.descriptionEn este estudio, se determinó el potencial celulolítico de cepas bacterianas asociadas al compost de estiércol bovino. Para esto, se estableció un método de cribado para la identificación de cepas bacterianas con actividad celulolítica basado en el uso de rojo congo. Se evaluó de manera semicuantitativa la actividad celulasa de 49 cepas termotolerantes provenientes de la Colección de Trabajo del Banco de Microorganismos de Sistemas Agropecuarios Sostenibles. Estas cepas fueron aisladas del compostaje de estiércol bovino y pasto. Para esto, se calcularon los índices celulolíticos basados en halos de degradación de carboximeticelulosa revelados con rojo congo y una solución salina. De igual forma, la evaluación cuantitativa de la actividad celulasa de dieciséis cepas termotolerantes de la colección, que obtuvieron un índice celulolítico mayor a 65% y 14 nuevas cepas termófilas, fue determinada de dos formas. En la primera, se estimó los azúcares formados en caldo 1% carboximetilcelulosa después de 48 horas de incubación. Finalmente, 27 cepas de la colección se caracterizaron basados en el 16S rRNA. El 74.04% de estas cepas se identificó como pertenecientes al género Bacillus, el 10.11% al género Ralstonia, el 7.40% al género Undibacterium, y el resto correspondieron a los géneros Klebsiella, Zeimonas y Priestia cada uno con 3.70%.
