This Standard Operating Procedure (SOP) describes a shotgun lipidomics method for comprehensive lipid analysis in plant and plant by-products foods implemented on a Sciex 5500 or 6500 instruments with Selexion Technology. The protocol covers extraction of 22 lipid classes from lyophilized plant material using a modified Bligh-Dyer method, followed by DI-DMS-MS/MS analysis with two complementary acquisition methods. The procedure includes quality control measures, DMS tuning optimization, and automated data processing through Shotgun Lipidomics Assistant software, designed specifically for the Periodic Table of Food Initiative (PTFI) to characterize lipid diversity across plant species.
